Теоретически, сок грейпфрута может увеличивать концентрацию в плазме крови лоратадина, как и других препаратов, которые являются субстратами CYP450 3А4 ферментативного пути. Предлагаемый механизм заключается в ингибировании CYP450 3А4-опосредованный метаболизм первого прохода в стенке кишечника путем определенного соединения, содержащиеся в грейпфрутах. Клиническое значение этих потенциальных взаимодействий неизвестны. Сообщается взаимодействия с мощными ингибиторами CYP450 3А4, такие как Кларитромицин, эритромицин и кетоконазол произвели существенное увеличение площади под плазменной концентрации время кривой (AUC) лоратадина и его активного метаболита, дескарбоэтоксилоратадин, без соответствующих изменений в общий профиль безопасности препарата.
